HPLC vs MS vs net peptide content: a decision tree for research peptide CoAs

Three fields drive most peptide receiving decisions. Confusing them is how labs accept incomplete documentation. Use this decision tree before releasing a lot into inventory.

Start with lot linkage

If vial lot ≠ CoA lot, stop. No purity number fixes a chain-of-custody break.

Branch A — HPLC purity present?

Yes: record method wavelength/conditions if provided and archive the chromatogram when available.

No: quarantine. Area-percent purity is a baseline release field for synthetic peptides.

Branch B — MS identity present?

Yes: confirm observed mass matches the expected free acid/amide/acylated form for the SKU.

No: quarantine for identity risk even if HPLC looks excellent.

Branch C — Will you calculate concentration?

If yes, require net peptide content. Gross mass alone biases molarity.

If no (qualitative presence only), still prefer net peptide for future assay reuse.

Branch D — Release or escalate

Release only when lot linkage, HPLC, and MS clear your SOP. Escalate to the supplier when any required field is missing or inconsistent.
